ABSTRACT

To study the interaction between C4b-binding protein (C4BP) and Riemerella anatipestifer (RA), we cloned duck C4BPα, conducted prokaryotic expression and prepared the polyclonal antibody by immunizing mice. Then indirect immunofluorescence assay and dot blotting hybridization assay were used to verify the interaction between C4BP and RA. The full length of duck C4BPα nucleotide sequence was 1 230 bp, with the highest similarity to chicken C4BPα (82.1%). Phylogenetic tree analysis showed that duck C4BPα and chicken C4BPα were on the same phylogenetic tree branch and the genetic evolution relationship between them was the closest. C4BPα was efficiently expressed in Escherichia coli BL21 (DE3). The recombinant proteins existed in intracellular soluble form. The titer of polyclonal antibody was more than 1:10 000 and polyclonal antibodies could specifically recognize the recombinant proteins. The results of indirect immunofluorescence assay and dot blot hybridization assay showed that RA could interact with duck C4BP. The results provide a basis to further reveal the pathogenesis of RA.

ABSTRACT

Objective@#To investigate the different patterns of 24-h movement behaviors, and their associations with sociodemographic factors in a nationally representative sample of Chinese children aged 6-13 years.@*Methods@#This study was based on a national multi-centered cluster intervention study involving 31 362 children aged 6-13 years from 7 provinces in China. Questionnaires were used to collect moderate to vigorous physical activity (MVPA), screen time (ST) and sleep duration, as well as sociodemographic variables including age, gender, area of residence, parents’ education level and family income. Generalized linear mixed model (GLMM) analyses were conducted for the 24-hour movement behaviors according to sociodemographic variables.@*Results@#The proportions of individuals meeting the MVPA, ST, and sleep guidelines were 32.2%, 78.5%, 30.1%, respectively. The proportion that meet 0, 1, 2 and 3 recommended items was 9.6%, 47.7%, 35.0% and 7.7%, respectively. Age, gender, parents’ education levels and family income showed associations with PA, ST and sleep. Compared with low parents’ education group, the risk for unhealthy behavioral patterns was lower in those with high parents’ education level(P<0.05).@*Conclusion@#The current status of 24-hour physical activity for children aged 6-13 in China is not ideal, and social demographics should be considered when designing targeted interventions to promote children’s health.

ABSTRACT

Objective To explore the association of urinary level of kidney injury molecule-1 (KIM-1) with renal tubulointerstitial lesions in patients with lupus nephritis (LN). And to find a potential clinical biomarker, which could reflect the specific tubulointerstitial lesions of LN. Methods Sixty cases of biopsy proven new-onset LN patients were enrolled into the study. And 10 normal kidney tissues were collected, which came from 10 patients with kidney trauma or renal tumor nephrectomy. Sixty patients with LN were divided into mild disease group, moderate disease group, severe disease group and the extremely severe disease group according to the criteria of 2000 Hill's morphologic index for the evaluation of renal pathology. Urine and renal tissues specimen were collected. The clinical and pathological data were analyzed retrospectively. Urine level of KIM-1 was detected by enzyme linked immunosorbent assay (-). Immunohistochemical staining was used to observe the expression of KIM-1 in the renal tissue. T-test, One-Way analysis of variance(ANOVA) test or rank sum test and Pearson or Spearman correlation analysis were used for statistical analysis. Results ①With the aggravation of tubulointerstitial lesions, the urinary level of KIM-1 was increased gradually, which was shown in the control group [(0.32 ±0.03) ng/ml], mild group [(2.31 ±0.30) ng/ml], moderate group [(6.12 ±0.51) ng/ml], severe group [(12.51 ±1.83) ng/ml] and the extremely severe group [(15.30 ±2.11) ng/ml] (all P0.05). The expression of KIM-1 in the renal tissue was positively correlated with serum creatinine (SCr), quantity of 24 h UP, SLEDAI, R-SLEDAI, GAI, TLAI and AI (r=0.780, 0.780, 0.812, 0.727, 0.735, 0.910, 0.701, P0.05). Conclusion Urinary KIM-1 enables to assess the tubulointerstitial lesion in LN patients, and it can be a sensitive biomarker to predict the tubulointerstitial damage and to evaluate the degree of tubulointerstitial lesions.

ABSTRACT

Objective To investigate the effect of myeloid differentiation factor88 (MyD88)on the experimental autoimmune myocarditis (EAM) in Balb/c mice.MethodsTotally 90 Balb/c mice were randomly divided into three groups:MyD88 ( n =30),myocarditis ( n =30),and control ( n =30 )groups.An injection of MyD88 high expression adenovirus was carried out through vena caudalis at day 3.The mice of myocarditis group received porcine cardiac myosin at the same time and position.The mice of control group received PBS only.Serum and myocardium samples were gained at day 21.The inflammatory infiltration and the serum levels of cTnI,IgG,and IgM were examined.Results 21 days later,the inflammatory infiltration score of the myocarditis group was higher than the control group (2.39 ± 1.23 vs 0.68 ±0.65,P <0.05).The MyD88 group was significantly higher than the other 2 groups ( F =94.194,P <0.01 ),the myocarditis group ( 3.56 ± 1.34 vs 2.39 ± 1.23,P < 0.05 ),and the control group ( 3.56 ±1.34 vs 0.68 ±0.65,P <0.01).The serum levels of cTnI,IgG,and IgM of the MyD88 group were significantly higher than the myocarditis group and the control group( P <0.01,P <0.05).Conclusions MyD88 participates in the pathogenesis of experimental autoimmune myocarditis in mice.

ABSTRACT

Objective To study the lipid regulatory effects of mannose ester and its effects on serum oxidized low density lipoprotein(OX-LDL),C-reactive protein(CRP)in patiends with coronary heart disease(CHD)complicated with hyperlipidemia.Methods Seventy-six CHD patients with hyperlipidemia were treated with mannose ester at a dose of 0.1g orally,three times a day for 2 successive months.Levels of blood lipids,OX-LDL,CRP at the time of entry,first month,second month of the therapeutic course were observed.Results Mannose ester could reduce the serum levels of total cholesterol,LDL-C 1 month after medication and the effect was further elevated after 2 months.Meanwhile its effects in lowering triglycerides and increasing high density lipoprotein were observed after 2 months administration.Mannose ester could also significantly lower the serum OX-LDL,CRP after 2 months administration.Conclusion Mannose ester has good regulatory effect on blood lipids,and decreases OX-LDL,CRP.Mannose ester can protect vascular endothelial cells and inhibit the development of inflammation in coronary plaque.Therefore,it can improve the prognosis of CHD patients.
